The Small RNA Sample Preparation kit is used to physically isolate small RNA, ligate the adapters necessary for use during cluster creation, and reverse-transcribe and PCR. Two protocols utilize this kit; Small RNA Sample Prep and Small RNA v1.5 Sample Prep.

  • The 5ʹ small RNA adapter is used in both protocols to amplify the small RNA fragment. This adapter also contains the DNA sequencing primer binding site.
  • The 3ʹ small RNA adapter needed only in the Small RNA sample prep protocol is used for reverse transcription and corresponds to the surface bound amplification primer on the flow cell used during cluster generation
  • The v1.5 small RNA 3ʹ adapter used only in the Small RNA v1.5 sample prep protocol is specifically modified to target microRNAs and other small RNAs that have a 3ʹ hydroxyl group resulting from enzymatic cleavage by Dicer or other RNA processing enzymes
  • Libraries prepared with the Small RNA Sample Preparation Kit can be sequenced on the HiSeq 2000, HiSeq 1000, HiScanSQ, or Genome Analyzer.
