Sequencing the DNA That Encodes Those Tiny Fingers and Toes

A team led by Jay Shendure, M.D., Ph.D., Associate Professor of Genome Sciences at the University of Washington, and doctoral candidate Jacob Kitzman, developed a haplotype-resolved genome sequencing method that combines the throughput of next-generation sequencing with the contiguity information provided by large-insert cloning. Recognizing its potential value in prenatal genetics, the team used the method and the HiSeq® 2000 system to determine the whole-genome sequences of two fetuses, one at 18 weeks and the other at 8 weeks gestation.

Dr. Martin Widschwendter and researchers at the University College of London found that age-dependent DNA methylation of certain stem cell genes may prime cells for malignant transformation, tipping the balance in favor of cancer.,It is widely recognized that a number of factors, such as exposure to smoking, chemicals, or steroids, can lead to cancer. One of the mechanisms that may be responsible is an alteration in DNA methylation (DNAm) patterns, resulting in the hypermethylation of tumor suppressor gene promoters. While this represents an important component, it is certainly not the lone trigger of cancer development. Martin Widschwendter, M.D., MRCOG and his team at the University College London’s Institute for Women’s Health investigated whether DNA methylation of genes targeted by polycomb group proteins (PCGs) in stem cells may also play a role in cancer.

Illumina's Genome Analyzer is shedding light on the genomic differences between bacterial isolates, enabling the creation of a pathogen database that will allow scientists to better track disease outbreaks. Stephen Bentley, Ph.D. and a team of scientists at the Sanger Institute decided to test the use of next-generation sequencing as a molecular typing platform to see if it could provide greater insight into the micro-evolutionary changes occurring in the bacterial genome. Their ultimate goal is to create a database with enough genomic clarity to be useful to clinicians and epidemiologists during local outbreaks and pandemics.
