Overview

The NovaSeq 6000 Xp Workflow supports a variety of applications, such as whole-genome sequencing (WGS), ultradeep exome sequencing, and single-cell sequencing. Improving upon the standard workflow, the NovaSeq 6000 Xp Workflow adds a level of flexibility and control to experimental planning and flow cell configuration on the NovaSeq 6000 System.

The NovaSeq 6000 Xp Workflow requires one of the following kits:

  • NovaSeq 6000 Xp 2-Lane Kit v1.5: Compatible with NovaSeq 6000 SP, S1, and S2 flow cells

  • NovaSeq 6000 Xp 4-Lane Kit v1.5: Compatible with NovaSeq 6000 S4 flow cells

These kits contain reagents and a disposable manifold for sample loading.  

 

The NovaSeq 6000 Xp Flow Cell Dock is a one-time purchase, reusable hardware accessory that is required for the NovaSeq 6000 Xp Workflow.

The NovaSeq 6000 Xp Workflow enables sequencing of different libraries (pools) in each lane of the NovaSeq flow cell. Additionally, the ExAmp/library mix in the Xp workflow is prepared manually and loaded onto the flow cell using the NovaSeq 6000 Xp Flow Cell Dock.

The NovaSeq 6000 Xp Workflow follows the same steps as the standard workflow until after library denaturing. Denatured libraries are manually added to the ExAmp master mix and directly loaded into individual lanes of the flow cell using the NovaSeq 6000 Xp flow cell dock. The filled flow cell is then loaded onto the instrument for clustering and sequencing.

The NovaSeq 6000 Xp kit is available in two sizes. The NovaSeq 6000 Xp 2-lane kit v1.5 is used with SP, S1, and S2 flow cells. The NovaSeq 6000 Xp 4-lane kit v1.5 is used with S4 flow cells. Each kit contains a single-use manifold and sufficient reagents for one NovaSeq flow cell.

The NovaSeq 6000 Xp Workflow allows for each flow cell lane to be loaded independently. This allows sequencing of different libraries in each lane of the flow cell.
