SMDB

SMDB is a method to leverage short-read sequencing to obtain long and accurate reads. Using a microfluidics system, the method isolates, amplifies, fragments, and barcodes single DNA molecules in aqueous picoliter droplets. This approach allows the full-length molecules to be sequenced with multi-fold coverage, using short-read sequencing.

Pros:
  • Limit of detection scales with the number of molecules sequenced and can be orders of magnitude more sensitive than conventional NGS
Cons:
  • Requires specialized hardware
